If you are using cement head glue it shouldn't take but an hour or so. If the thread is coming untied it sounds like it is old, or someone didn't know how to tie a whip finish knot.

I not a fly rod enthusiast, I have a couple ,but haven't picked them up in years. Anyway I have certain lures that I per-sue trout with and after a few trout i notice the thread coming unraveled. Causing the feather or what ever it is to pull out and lose it's appeal. My friend who ties flies told me to get some glue from the fly shop. So I purchased some "fly head glue"? That's what the guy recommended. Said it would soak into the thread. My question to you guys is how long does it take for this stuff to set up? Hope I make sense. and thanks

Yeah cement head glue that's what I got. good help is getting hard to find! although those sand paper lips on a trout take a toll on a lure.
While cement head glue is the correct thing to use ,in a pinch one can use clear fingernail polish.
